چکیده

Boiled silkworm pupa is a traditional food in Asia, and patients with silkworm pupa food allergy are common in these regions. Still now only one allergen from silkworm, arginine kinase, has been identified. The purpose of this study was to identify novel food allergens in silkworm pupa by analyzing a protein extract after heat treatment. Heat treated extracts were examined by proteomic analysis. A 27-kDa glycoprotein was identified, expressed in Escherichia coli, and purified. IgE reactivity of the recombinant protein was investigated by ELISA. High molecular weight proteins (above 100 kDa) elicited increased IgE binding after heat treatment compared to that before heat treatment. The molecular identities of these proteins, however, could not be determined. IgE reactivity toward a 27-kDa glycoprotein was also increased after heating the protein extract. The recombinant protein was recognized by IgE antibodies from allergic subjects (33.3%). Glycation or aggregation of protein by heating may create new IgE binding epitopes. Heat stable allergens are shown to be important in silkworm allergy. Sensitization to the 27-kDa glycoprotein from silkworm may contribute to elevation of IgE to silkworm.

Identification and characterization of an arginine kinase as a major allergen from silkworm (Bombyx mori) larvae.

BACKGROUND The silkworm, Bombyx mori, is an important insect in the textile industry and its pupa are used in Chinese cuisine and traditional Chinese medicine. The silk, urine and dander of silkworms is often the cause of allergies in sericulture workers and the pupa has been found to be a food allergen in China. Recent studies have focused on reporting cases of silkworm allergies, but only a f...

The silk proteins, sericin and fibroin in silkworm, Bombyx mori Linn., - a review

The domesticated silkworm, Bombyx mori Linn., a lepidopteran molecular model and an important economic insect that are emerging as an ideal molecular genetic resource for solving a broad range of biological problems. The silkworm, B. mori produces massive amount of silk proteins during the final stage of larval development. These proteins are stored in the middle silk gland and they are dischar...

Effect of 12-hours delay and anticipation from traditional practice in silkworm mounting to spinning frames on economic traits of Bombyx mori (Lepidoptera: Bombycidae) cocoons

In this study, the aim was to determine the effect of 12 hours delay and time advancement of Bombyx mori larvae transfer to spinning frames on the economic traits of harvested cocoons. Our experience focused on the transfer of three lots of silkworm mature larvae to cocoon frames with 12 hours delay or advancement from usual spinning time. Commercial silkworm eggs were obtained and thr...

Proteomic and immunological identification of two new allergens from silkworm (Bombyx mori L.) pupae

This study explored food allergy caused by eating silkworm (Bombyx mori L.) pupae, a traditionally accepted food and animal feed in East and Southeast Asia, and identified two new allergens by proteomic and immunological methods. Proteins isolated from silkworm pupae were separated by two-dimensional gel electrophoresis (2-DE); pooled sera from patients allergic to silkworm pupa proteins were u...

Molecular Characterization, Tissue Distribution, Subcellular Localization and Actin-Sequestering Function of a Thymosin Protein from Silkworm

We identified a novel gene encoding a Bombyx mori thymosin (BmTHY) protein from a cDNA library of silkworm pupae, which has an open reading frame (ORF) of 399 bp encoding 132 amino acids. It was found by bioinformatics that BmTHY gene consisted of three exons and two introns and BmTHY was highly homologous to thymosin betas (Tβ). BmTHY has a conserved motif LKHTET with only one amino acid diffe...
